Description:
Lynparza is an inhibitor of poly (ADP-ribose) polymerase (PARP) enzymes, including PARP1, PARP2, and PARP3. PARP enzymes are involved in normal cellular homeostasis, such as DNA transcription, cell cycle regulation, and DNA repair. Olaparib has been shown to inhibit growth of select tumor cell lines in vitro and decrease tumor growth in mouse xenograft models of human cancer both as monotherapy or following platinum-based chemotherapy.
Increased cytotoxicity and anti-tumor activity following treatment with olaparib were noted in cell lines and mouse tumor models with deficiencies in BRCA. In vitro studies have shown that olaparib-induced cytotoxicity may involve inhibition of PARP enzymatic activity and increased formation of PARP-DNA complex, resulting in disruption of cellular homeostasis and cell death.

Do NOT use olaparib if:
you are allergic to any ingredient in olaparib
you still have blood problems caused by previous chemotherapy treatments
you are taking carbamazepine, phenytoin, rifampin, or St. John's wort

How to use olaparib:
Use olaparib as directed by your doctor. Check the label on the medicine for exact dosing instructions.
Olaparib comes with an extra patient information sheet called a Medication Guide. Read it carefully. Read it again each time you get olaparib refilled.
Swallow olaparib whole. Do not chew, dissolve, or open the capsule before swallowing.
Do not take the capsule if it is deformed or leaking.
Avoid food or drink containing grapefruit or Seville oranges while you are taking olaparib.
Continue to take olaparib even if you feel well. Do not miss any doses.
If you miss a dose of olaparib, skip the missed dose and go back to your regular dosing schedule. Do not take 2 doses at once.
